Black yeast Aureobasidium pullulans extracellularly produces β-(1,3),(1,6)-D-glucan under certain conditions. In this study, using Glycine max cv. Kurosengoku (Kurosengoku soybeans), the production of β-glucan through fermentation of A. pullulans was evaluated, and the effects of A. pullulans cultured fluid (AP-CF) containing β-glucan made with Kurosengoku soybeans (kAP-CF) on human monocyte derived Mono Mac 6 cells were investigated. An anti-angiogenic protein, Thrombospondin-1 (THBS1) was effectively induced after stimulation with kAP-CF. The combined stimulation of β-glucan with Kurosengoku soybean extract more effectively induced THBS1 than soybean extract alone. These results suggest synergistic effects of A. pullulans-produced β-glucan on the enhancement of Kurosengoku soybean-induced THBS1 expression.
This study reveals a novel synergistic mechanism by which Aureobasidium pullulans beta-glucan enhances its anti-cancer potential through combination with Kurosengoku (black soybean) extract — specifically by upregulating thrombospondin-1 (TSP-1), the body's own anti-angiogenic protein.
Key findings:
Significance: Angiogenesis — the formation of new blood vessels to supply growing tumors — is a hallmark of cancer progression. TSP-1 is one of the body's natural defenses against this process. This study shows AP-beta-glucan can amplify TSP-1 production, especially in combination with polyphenol-rich Kurosengoku soybean. This is a distinct anti-cancer mechanism from NK cell activation (the most-studied AP-glucan pathway), adding a third mode of anti-tumor action: direct apoptosis, NK cell activation, AND anti-angiogenesis via TSP-1 upregulation.
