Poliovirus type 1 mutants were created in stem-loop II of the internal ribosome entry site. Two deletion mutants showed different defects: one impaired both genome RNA synthesis and viral protein production, while the other mainly impaired protein production. Differences in host-protein binding supported overlapping RNA-replication and translation functions in this region.
